Uses of Adenafil 100mg

Adenafil 100 contains the main ingredient is sildenafil citrate, used to treat erectile dysfunction but requires sexual stimulation to be effective. 

1. The function of Adenafil 100mg

Adenafil is used to treat erectile dysfunction to improve, achieve or maintain an erection sufficient for satisfactory sexual activity. The drug is only effective when accompanied by sexual stimulation. However, Adenafil also has some contraindications as follows:

  • Children and women.
  • Patients with medical history of coronary or cerebrovascular disease.
  • Patients are allergic with any ingredients of medication.
  • As sildenafil enhances the hypotensive effect of acute and chronic nitrate so patients who are using organic nitrate salts regularly or intermittently are contraindicated to use this medication;
  • The patient lost vision in one eye due to non-arteritic anterior ischemic optic neuropathy (NAION).

2. How to use Adenafil?

For adult men, most patients are recommended to use 50mg, take it 1h before having sex, maximum dose is 100mg/day. Patients with kidney failure should use these following reference doses:

  • Mild to moderate kidney failure (creatinine clearance > 30 - 80 ml/min): do not need to adjust dosage.
  • Severe kidney failure (creatinine clearance < 30 ml/min):  use 25mg since the clearance of sildenafil  is reduced in these patients.
  • Patients with liver failure: use 25mg since the clearance of sildenafil  is reduced in these patients.

3. Side effects of Adenafil

Use Adenafil may lead to undesired side effects such as:

  • Headache, dizziness, astigmatism;
  • Nausea, indigestion;
  • Eye pain, photophobia, dizziness;
  • Hyperemia, glare;
  • Tachycardia, palpitations;
  • Hypotension;
  • Rash;
  • Seizures, syncope;
  • Involuntary erection

In terms of medication interaction, Sildenafil is  metabolized mainly via cytochrome P450 3A4 and 2B9, so the medication inhibit cytochrome such as Cimetidine, Erythromycin, Ketoconazol, Itraconazol, Ritonavir, Saquinavir,... will reduce the clearance of  Sildenavir thus leading to increased plasma concentrations in plasma. When Sildenafil is used concurrently with cytochrome inducers, its plasma concentration decreases. Antacids such as magnesium hydroxide and aluminum hydroxide do not affect the bioavailability of Sildenafil citrate.
