EXOSOMES DERIVED FROM HUMAN INDUCED PLURIPOTENT STEM CELLS AMELIORATE THE AGING OF SKIN FIBROBLASTS

This study reported the effect of exosomes (EX) from the conditioned medium of human induced pluripotent stem cells (iPSCs) on aged human dermal fibroblasts (HDFs).

Materials:

- human iPSC line was generated from human dermal fibroblasts by introducing Oct4, Sox2, cMyc, and Klf4 using Sendai Virus.

- Irradiated HDMs were created by UV irradiation.

Methods:

  • EXs were collected using ExoQuick-TC TM
  • Cell proliferation and migration assay

Results:

  • The proliferation of HDFs was increased by iPSC-Exo in a dose-dependent manner up to 50 × 10 8 particles/mL and maintained with a higher concentration
  • iPSC-Exo promoted the migration of HDFs
  • HDMs were treated with 20 × 10 8 iPSC-EX prior to UVB irradiation. The iPSC-Exo-treated group showed viabilities of 88.4% ± 2.1%, while the viabilities of the untreated group were significantly reduced to 65.6% ± 8.6%.
  • iPSC-EX treatment significantly increased transcript levels of collagen type I, which is a crucial component of the skin dermis. On the contrary, the expression levels of matrix-degrading enzymes (MMP-1 and MMP-3) were decreased in the HDFs treated with iPSC-EX (The high level of MMP-1 and MMP-3 were observed in photoaging cells).
  • iPSC-EX treatment significantly decreased transcript levels of SA-β-Gal. The SA-β-Gal is a marker of senescence of cells. This indicates iPSC-EX can protect cell from aging.

The results from this study indicate that iPSC-EX can enhance the HDF proliferation and migration. In addition, the results demonstrated that iPSCs-Exo significantly reduced the expression level of SA-β-Gal and MMP-1/3 and restored the collagen type I expression in senescent HDFs. Taken together, it is anticipated that these results suggest a therapeutic potential of iPSCs-Exo for the treatment of skin aging.
