2. What is a CSF culture?


CSF culture is to put the cerebrospinal fluid (or CSF) into a suitable environment with sufficient nutrients, then incubated at 37oC, CO2 (5%) to create conditions for bacteria (if necessary). yes) is developed. 3. How to get CSF culture specimens


Lumbar puncture is currently the safest and most common method for obtaining cerebrospinal fluid. The person performing the procedure must be a qualified physician who is well trained in the puncture procedure. 4. Cerebrospinal fluid culture procedure


At the laboratory:
Direct observation of cerebrospinal fluid: clear or cloudy, yellow or milky? Observation can guide the cause. Direct endoscopic staining from specimens: endoscopic staining after natural sedimentation, or after centrifugation. Gram stain to observe white blood cells, observe bacteria. The orientation of cocci, bacilli, gram-negative or gram-positive will help the clinician to use antibiotics when the results of antibiogram are not available. If TB is suspected, stain with Zielh Neelsen. Inform the clinician about the results of endoscopic staining. Culture: Use a sterile pipette to aspirate CSF and add 2-3 drops to blood agar, chocolate agar, UTI agar. Use sterile cultures to separate regions to create separate colonies. Bacterial proliferation is possible in BHI medium. Incubate blood agar and chocolate agar at 37oC, 5% CO2, incubate UTI agar at 37oC in normal atmosphere. After 24 hours, read the results: observe the colonies growing on the agar media and subculture from BHI, staining from the colonies. Then identify the bacteria and make an antibiogram. In case after 24h, bacteria do not grow, further incubate, and observe daily up to 72h. Returns negative if no growth.
